The main components of the endomembrane system are the ions, amino acids, ATP, and other small molecules that randomly diffuse throughout the cell.  The secretory pathway hypothesis is that such a pathway exists and that the rough endoplasmic reticulum and Golgi apparatus work together as an integrated endomembrane system.    The secretory pathway hypothesis experimentally was confirmed because it suggested proteins that were specified for secretion from the cell that gets synthesized in a particular set of steps. These proteins are placed into vesicles when moving from the rough endoplasmic reticulum to the Golgi Apparatus.  The rough endoplasmic reticulum and the Golgi apparatus work together as an integrated endomembrane system.    The "pulse- chase" experiment proves that a labeled molecule is followed by a chase molecule. Thus, proteins can be tracked.
