The sentence with the comma splice is number 1. You can't join two independent clauses with a comma. In more informal writing, comma splices can be used, but this is academic writing.

Question:

What is a theme that stood out to you from our text, "The Joy Luck Club" by Amy Tan and why?. (Please remember to support your answer with evidence from the text!)

Answer:

One of the focal themes for "The Joy Luck Club" for me was the theme of cultural identity. All of the moms were born in China and all of the daughters were born in America. There's a disconnect between generations that causes misunderstandings in expectations and the importance of heritage. The clash of culture between mother and daughter can be tumultuous. It stood out to me because I thought that the daughters were in the right, but heritage and culture from previous generations is deeply important to who we are. In ignoring or disliking our parents' heritage and history, we deny ourselves important experiences in life.
